Determination and Study on Pharmacokinetics of Metronidazole and Tinidazole in Human Breast Milk

  • 摘要: 采用高效液相色谱法测定哺乳妇女单剂量静滴甲硝唑(20mg/kg,n=8)、替硝唑(13mg/kg,n=7)乳汁药物浓度,色谱柱为pecoshere C18(3μm,3.3cm×4.6mm),氯仿提取乳汁中药物,其平均回收率大于88.8%.乳汁中甲硝唑与替硝唑药代动力学参数tmax,Cmax,t1/2Ke分别为1.7±1.0h,20.10±4.95μg/mL,6.4±3.3h和1.3±0.6h,17.23±3.12μg/mL,11.0±3.5h.

     

    Abstract: The metronidazole and tinidazole in human breast milk were assayed by high performance liquid chromatography(HPLC). A high performance and rapid column pecosphere C18 (3 μm, 3.3 cm×4.6 mm) was used. Metronidazole and tinidazole in breast milk extracted with chloroform, the average recovery rates from breast milk were above 88.8%. The pharmacokinetic parameters(mean±s) tmax,Cmax,t1/2Ke of Metronidazole and Tinidazole in lactating women after infusion of a single dose of Metronidazole(20 mg/kg, n=8) and Tinidazole (13 mg/kg, n=7) respectively were 1.7±1.0 h, 20.10±4.95 μg/mL, 6.4±3.3 h and 1.3±0.6 h, 17.23±3.12 μg/mL, 11.0±3.5 h.
