Study on the Analysis of Chlorogenic Acid in Tobacco

  • 摘要: 采用超声波提取、反相高效液相色谱测定烟叶中绿原酸,用内标法进行定量,并对超声波提取烟叶中绿原酸的提取条件进行了优化,结果表明:用60%的乙醇溶液,在50℃、pH 4条件下,超声提取45 min效果最佳.该法具有良好的线性(相关系数为0.999 9),检测限为1.4×10-2 mg/10 mL(S/N=3)加标回收率为101.5%,方法RSD为1.58%.方法快速、简单、准确.

     

    Abstract: A simple and rapid method has been developed for determination of chlorogenic acid in tobacco by RP-HPLC and the extracting process was optimized. The quantitative results were obtained by the internal standard method. The results indicated that the optimum extraction process is as follows: the content of ethanol is 60%,the temperature of ultrasound is 50℃,the pH of ethanol solution is 4, the time of extraction is 45 minutes. The method has good linearity with correlation coefficient of 0.9999.The limit of detection based on signal-to-noise ratio of 3 was 1.4×10-2 mg/10 mL. The recovery of chlorogenic acid in tobacco was 101.5%,with RSD of 1.58%.
