高效液相色谱法测定菌肥中赤霉素和吲哚乙酸含量方法研究

Determination of the Content of Gibberellin A3 and Indoleacetic Acid from Fertilizer by HPLC

  • 摘要: 建立了一种高效液相色谱法分离测定赤霉素(GA3)和吲哚乙酸(IAA)的方法.采用Agilent ZORBA×300SB-C18(4.6 mm×250 mm)色谱柱,以甲醇:水=35:65,(V/V,含2%冰醋酸)为流动相,柱温为25℃,流速为0.5 mL/min.系统讨论了流动相组成及pH对分离的影响.本法测定GA3和IAA的线性范围分别为25.0~1 000μg/mL、2.5~50.0μg/mL.方法回收率分别为85.63%、90.21%.

     

    Abstract: To develop a reliable method for the determination of the content of Gibberellin A3 and Indoleacetic acid by HPLC,an Agilent ZORBA×300SB-C18(4.6 mm×250 mm)column was used.The mobile phase was CH3OH:H2O=35:65(V/V,2% CH3COOH) at a flow rate of 0.5 mL/min,the column temperature was 25℃,and an IR detector was used.The content of samples were quantitated by peak area with external standard.The standard curves of GA3 and IAA were linear in the range of 25.0~1 000 μg/mL and 2.5~50.0 μg/mL,respectively.The average recovery were 85.63% and 90.21,respectively.The effects of several HPLC parameters on the resolution were studied systematically.The results demonstrate that the proposed method also promises to be applicable to the quality control of the fertilizer.
