Determination of Fanchinoline and Tetrandrine in Plasma of rats by HPLC

  • 摘要: 建立了高效液相色谱(HPLC)快速测定血浆中2种生物活性碱-防己诺林碱和粉防己碱的方法.血浆经乙腈沉淀蛋白,稀释后直接进样,采用ODS C18柱,乙腈-磷酸缓冲液-三乙胺(80∶30∶0.3,V/V/V)为流动相,紫外检测波长282 nm,外标法定量.为研究防己诺林碱和粉防己碱在生物体内的药物代谢机理研究提供了简便、灵敏、准确的分离测定方法.

     

    Abstract: High Performance Liquid Chromatographic (HPLC) method was employed for the determination of fanchinoline and tetrandrine in plasma of rats. The plasma sample was precipitated with acetonitrile and the supernatant was diluted and then directly injected into ODS C18 column with acetonitrile-phosphate buffer-triethylamine (80∶30∶0.3,V/V/V) as the mobile phase. UV detection was set at 282 nm and standard curve was for the quantification. This assay could be used for separation and determination of fanchinoline and tetrandrine in the bioactivity organism simply and accurately.
