The Study on the Hydrolysis Rate Constants of Daphnin by Capillary Electrophoresis

  • 摘要: 采用毛细管电泳-紫外检测法对瑞香苷进行了水解动力学研究.瑞香素是瑞香苷的水解产物. 考察了瑞香苷(DN)和瑞香素(DP)的紫外吸收随水解反应过程的进行而发生的变化,计算得水解的速率常数,并总结了酸度和温度对速率常数的影响规律.当盐酸浓度为1.0 mol/L,温度为333、347、351、363、371 K时的水解常数分别为1.1×10-3、4.6×10-3、6.6×10-3、17.3×10-3、33.5×10-3 min-1. 同时,根据阿仑尼乌斯公式计算该反应的活化能为91.62 kJ/mol.此法用于测定瑞香苷水解常数简便、直观.

     

    Abstract: A high sensitivity and good repeatability method based on capillary electrophoresis with UV detection (CE-UV) was developed for kinesics study on the hydrolysis reaction of daphnin. Daphnetin is the product of the hydrolysis reaction of daphnin. The changes of ultraviolet absorptions of both daphnin and daphnetin during the process of hydrolysis were described, and the hydrolysis rate constant was calculated, also the influence of temperature and acidity was summarized. Under the condition of 1.0 mol/L HCl as the reaction medium, at 333, 347, 351,363 and 371 K, the measured rate constants of daphnin hydrolysis were 1.1×10-3,4.6×10-3,6.6×10-3,17.3×10-3 and 33.5×10-3 min-1, respectively. The activation energy for daphnin hydrolysis was calculated to be 91.62 kJ/mol.
