HPLC Determination of the Organic Acid Content inFermentation Liquor of In-situ-biological Process for Oil Production

  • Using duality mobile phase of sodium dihydrogenphosphate buffer solution (pH=2.3) and acetonitrile (96∶4,V/V), we could determine the content of organic acid in fermentation liquor by reversed\|phase high performance liquid chromatographic method. Flow rate of mobile phase was 1.0 mL/min with UV-detection at 210 nm.The results showed that while the contents of organic acids were in the range of 0.05~1.5 g/L,the area and the content were in a well linear correlation(r≥0.9990,RSD<4.0%(n=8)), the average recovery was 100%±4%. This method also had the simple, rapid, well accuracy.
