Determination of Bergenin in Saxifragaceae Plant by High Performance Capillary Electrophoresis

  • 摘要: 以苯乙胺为内标,建立了高效毛细管电泳法(HPCE)测定虎耳草属植物中虎耳草素含量的方法.讨论了缓冲溶液体系、pH值及缓冲液浓度对分离的影响.结果表明,以10 mmol/L硼砂盐酸缓冲液(pH 9.00)为分离介质,虎耳草素标准品与内标实现基线分离,且实验结果线性关系良好(r=0.9989),日内和日间相对标准偏差分别为1.7%和4.6%.

     

    Abstract: A method, in which Benzene ethamine was used as the internal standard, was developed for the determination of bergenin in Saxifragaceae plant by high performance capillary electrophoresis. The effects of buffer system, pH value and the concentration of buffer were discussed. The bergenin and the internal standard were baseline separation with 10 mmol/L borate buffer at pH 9.00. The calibration curves exhibited good linearity between the ratios of the concentrations of bergenin and internal standard and the ratios of the peak area of bergenin and internal standard over the concentration range from 5 to 100 g/L with the correlation coefficients of 0.998 9. The relative standard deviations of the intra-day and inter-day were 1.7% and 4.6%, respectively.
