Different Pore Diameter Silica-based Polymer-bonded Phases for Separation of Proteins, Polypeptids and Amino-acid

  • 摘要: 使用7μm的单分散不同孔径(10nm、100nm)的硅酸为基体,表面键合共聚有机物层,并在此类键合相上考察了蛋白质、多肽、氨基酸分离的色谱性能,实验结果表明,梯度冲洗下可得到胃蛋白酶、核糖核酸酶、溶菌酶、糜蛋白酶源、孵清蛋白的快速而良好的分离;在3min内获得两种结构相似氨基酸的基线分离.分析速度快、效率高,此类柱填料具有推广应用的价值.

     

    Abstract: The preparation of macroporous silica matrices of different pore diameter, the bonding of polymer to 7μm different pore diameter silica for the preparation of a novel packing has been studied. The chromatographic characteristics of these packing were examined. The results showed that under gradient elution, Proteins (pepsin,ribonuclease, lysozyme, chymotrypsinogen, ovalbumin)were separated within 15min successfully. Similar structure Amino-acid were separated on base line less than 3min.It is suggested that these packings separate biopolymer quickly with high resolution, and they have a wide-range applied prospective.
