毛细管电泳-柱端安培检测测定莲子心中荷叶碱、芦丁和金丝桃甙的含量

Determination of Nuciferine, Rutin and Hyperoside in PlumulaNelumbinis by Capillary Electrophoresis with End-column Amperometric Detection

  • 摘要: 采用毛细管电泳-柱端安培检测测定莲子心中荷叶碱、芦丁和金丝桃甙的含量.研究了检测电位、运行缓冲液浓度和pH值, 分离电压和进样时间对分离和检测的影响.以微碳圆盘电极(Ф=0.5mm)为工作电极,检测电位为 +0.95 V ( vs.Ag/AgCl ),pH为7.25 的50 mmol/L Na2B4O7和100 mmol/L NaH2PO4缓冲液为运行液,当分离电压为15 kV时, 3种分析物在15 min 内完全分离.荷叶碱、芦丁和金丝桃甙的检出限(S/N=3)分别为0.02 μg/mL、0.05 μg/mL 和0.04 μg/mL.该方法已成功地应用于莲子心中上述3种活性成分的测定.

     

    Abstract: A method based on capillary electrophoresis with end-column amperometric detection (CE-AD) was developed for the determination of Nuciferine, Rutin and Hyperoside in Plumula Nelumbinis. The working electrode was a 500 μm diameter carbon disc electrode positioned opposite the outlet of capillary. The pH and concentration of running buffer, separation voltage, injection time and detection potential were investigated to acquire the optimum conditions. The three analytes could be well separated within 15 min in a 60 cm length capillary ( 25 μm×370 μm) at a separation voltage of 15 kV in a 50 mmoL/L Na2B4O7-100 mmoL/L NaH2PO4 running buffer (pH=7.25). The detections limit for Nuciferine, Rutin and Hyperoside is 0.02 μg/mL, 0.05 μg/mL and 0.04 μg/mL, respectively. The optimized CE-AD method was employed to analyze Nuciferine, Rutin and Hyperoside in Plumula Nelumbinis, and the assay results are satisfactory.
